In this on-demand webcast, experts review current standards and emerging evidence for HER2 testing and HER2-directed therapy in advanced gastroesophageal adenocarcinoma and biliary tract cancer, with emphasis on how biomarker results should guide treatment selection and sequencing. Experts also discuss guideline-based HER2 assessment, clinical implications of intratumoral HER2 heterogeneity and loss of HER2 expression after treatment, and the rationale for reassessing HER2 status at progression. Pivotal and emerging clinical trial data for trastuzumab-, zanidatamab-, and trastuzumab deruxtecan–based strategies are summarized, and faculty address how efficacy, PD-L1 status, prior therapy, comorbidities, and toxicity profiles may influence treatment decisions.
